Q: Can I pour this over a polyurethane rubber part or would it stick?

It might not stick tight, but the tear strength of silicone might be exceeded if it sticks at all. When I am doing two part molds of, for instance, a seashell, I use vaseline on the first half surface so that the second half pouring doesn't stick to the first half. You could consider light painting with vaseline, or even spraying your object with "Pam."

Q: I've been searching for a a way to craft a 3d life-like mold for a peach. Life-sized using resin. will this work?

This silicone captures very fine detail, so it would work well for what you want. However, you would have to do a two part mold to cast an entire peach, which could be tricky if you're new to mold making. You also may need to create a more rigid mother-mold around the silicone mold to keep the mold from deforming when making your production cast. You can find plenty of videos online about advanced mold making processes.

Should work as long as you properly coat the resin so that the silicone no longer comes in contact with the resin.

While mixing it's not working time. Working time refers to just that. After mixing that's how much time you have to work with it.
